Recent Treatment for Colon Cancer.

Cancer treatment is a highly specialized process. The treatment choice is often influenced by various factors such as the type of cancer, age, stage, site, the physical condition of the patient, symptoms etc. With such a significant number of factors to consider, the diagnosis needs to be specialized in a particular way, comprehensive and co-ordinated to arrive at the correct treatment plan.

For the past 50 years, oncologists have had fundamental medications available, agents, that can induce tumor reactions in patients with colorectal cancer. However, in cases of advanced colorectal cancer, these regimens are never healing.  The recently introduced concept of cancer stem cells (SCs) drive tumor growth suggests a reason for these therapeutic failures.

Current chemotherapeutics target quickly dividing cells but cancer SCs divide only slowly, and, they are moderately resistant to cytotoxic fundamental therapies. It also proposes a solution—development of therapeutics that target cancer SCs. However, there is a scarcity of information about the mechanisms by which SC populations are maintained and about the mechanisms by which tumor SCs are associated with colon cancer development. In this article, we talk about the mechanisms and recent developments in the identification and isolation of colon cancer SCs utilizing new SC markers.

We then discuss the role of SCs in homeostasis of the typical colonic epithelium and mechanisms by which dysregulation of crypt mechanisms can lead to the initiation and progression of colon cancer. Our hypothesis, which has received recent experimental support, is that the mechanism that joins variations from the gene level (eg, APC mutations) and abnormalities at the tissue level (eg, dysplasia, proliferative shift, and carcinoma) from cancer initiation to metastasis is SC overpopulation. Finally, we talk about the concept that symmetric cancer SC division is a primary mechanism that drives tumor growth and that development of a new generation of therapeutics that target colon cancer SCs by inhibiting symmetric SC division holds promise for truly curative approaches for patients with advanced colorectal cancers.
